Skin and its appendages lesions with fungal infection remain one of the most important problems in dermatology. The paper presents epidemiological data on fungal infections (e.g. onychomycosis) and discusses the problem of hepatotoxicity of antifungal drugs. Drugs with highest and lowest risk of adverse events are listed. Metabolism of antifungal drugs with cytochrome P450 system is reviewed with focus on Itraconazole. Particular attention is paid to hepatoprotective effect of Taurine (product of sulfur containing amino acids metabolism, Dibikor, PIK-PHARMA, Russia).
The paper presets results of the study of Taurine hepatoprotective efficacy in 104 patients with onychomycosis, taking antifungal drug itraconazole for a long time. Also hepatoprotective efficacy was compared to herbal extract of milk thistle. Electrochemical studies of itraconazole's and hepatoprotectors' influence on CYR450 3A4 activity were conducted.
Key words: hepatotoxicity, antifungals, Taurinum, Itraconazole, onychomycosis, hepatoprotectors, cytochrome P450 system, Dibikor, skin fungal infection.
